JERUSALEM, May 03, 2023 (GLOBE NEWSWIRE) — Entera Bio Ltd. (NASDAQ: ENTX) (“Entera” or the “Company”) a leader in the development of orally delivered peptides and therapeutic proteins, today announced that its pre-clinical manuscript entitled “Oral Delivery Technology Enabling Gastro-Mucosal Absorption of Glucagon-Like-Peptide-2 Analog (GLP-2), Teduglutide – A Novel Approach for Injection-Free Treatment of Short Bowel Syndrome” has been accepted for publication by the International Journal of Peptide Research and Therapeutics.

Short bowel syndrome (SBS) is a rare and potentially life-threatening malabsorptive condition caused by a significant loss of functional bowel mass (secondary to congenital defects or disease-associated loss of absorption) or physical bowel mass (secondary to extensive intestinal resection). SBS patients have a reduced ability to absorb nutrients and fluids and are at risk of malnutrition, unintended weight loss and additional symptoms due to the loss of essential vitamins and minerals1. SBS is the most common cause of chronic intestinal failure, accounting for approximately 75% of cases of chronic intestinal failure in adults and 50% such events in children.2 Teduglutide, a GLP-2 analog and mainstay treatment for SBS, is known to enhance intestinal absorption. Teduglutide treatment requires daily sub-cutaneous injections.

“There are several new generation GLP-2 analogs under development which may also require chronic injections. As part of our R&D initiatives, Entera is leveraging on its proprietary platform to enable a potential first-in-class oral GLP-2, daily tablet treatment for patients with short bowel syndrome (SBS) and other disorders requiring parenteral nutrition. We are looking forward to sharing our in vivo findings with the community in the coming weeks,” said Miranda Toledano, Chief Executive Officer of Entera.

About Entera Bio

Entera is a leader in the development of orally delivered macromolecules, including peptides and other therapeutic proteins. The Company focuses on significant unmet medical needs where a daily mini tablet form of a peptide treatment or replacement therapy holds the potential to transform the standard of care. The Company’s most advanced product candidates, EB613 for the treatment of high risk, post-menopausal osteoporosis and EB612 for the treatment of hypoparathyroidism, are in clinical development. Both EB612 and EB613 are being developed to address undertreated diseases which disproportionately afflict women. EB613 is the first oral, once daily mini tablet presentation of synthetic hPTH (1-34), (teriparatide), consisting of the exact same 34 amino acid sequence as daily subcutaneous teriparatide injection, Forteo®, a mainstay anabolic treatment which requires daily SC injections. A placebo controlled, dose ranging Phase 2 study of EB613 tablets (n= 161) met primary (PD/biomarker) and secondary endpoints (BMD) in a dose dependent manner and was presented at the ASBMR 2021 Annual Conference. A phase 1 PK study of novel PTH formulations is planned for H1 2023 to ascertain feasibility of a new hypo candidate (a prior formulation had positive Phase 2a data announced in 2015 and published in JBMR 2019) and for another potential indication. JERUSALEM, Sept. 12, 2022 (GLOBE NEWSWIRE) — Entera Bio Ltd. (NASDAQ: ENTX), (“Entera” or the “Company”) a leader in the development of orally delivered peptides and therapeutic proteins, today announced the details of “A Six-month Phase 2 Study of Oral PTH (EBP05) in Postmenopausal Women with Low Bone Mass – Dose Proportional Absorption and Effect on Lumbar Spine BMD (SUN-591)” poster presentation at the American Society for Bone and Mineral Research (ASBMR) Annual Meeting 2022, being held from September 9-12th in Austin, Texas. The full text of the abstract will be published to the ASBMR meeting website, and the e-poster will be viewable on Entera’s website as of Sunday September 11th as of 1pm CT.

The correlation results were derived from Entera’s six-month phase 2 Study of lead clinical candidate, EB613 (EBP05), the first daily oral hPTH (1-34) teriparatide formulation in 161 post-menopausal women with low bone mass. The primary endpoint of change in P1NP at 3 months and secondary endpoints of change in lumbar spine (LS), total hip and femoral neck Bone Mineral Density (BMD) were met, as previously reported (ASBMR 2021 poster FRI-237 and oral presentation LB-1116). The current abstract and poster presentation reveals excellent correlations between the dose of the oral formulation of EB613 and teriparatide hPTH(1-34) plasma concentrations at the 15 min time point (R =0.996) and a linear dose response for the change in lumbar spine BMD after 6 months of treatment (R =0.998).

“These new analyses of blood hPTH concentration shortly after a dose of oral EB613 tablets confirm a strong, statistically significant correlation between mean blood level and the dose of EB613 taken. This finding is consistent with excellent correlation between change in lumbar spine BMD and dose of EB613 after 6 months of treatment,” Said Dr. Arthur Santora, Chief Medical Officer at Entera.

“We are encouraged by this important correlation analysis from our positive phase 2 study of EB613. Recent debates at the ASBMR annual conference highlight the importance of potentially earlier intervention with osteoanabolic agents capable of repairing bone structure and increasing mass; versus initial treatment with anti-resorptive drugs that mostly stop bone loss. EB613 has unique potential in the osteoporosis treatment paradigm as the first convenient, daily tablets form of osteoanabolic treatment,” said Miranda Toledano, Chief Executive Officer of Entera.

About EB613 (a.k.a. EBP05)

Parathyroid hormone (PTH) is an 84-amino acid hormone and the primary regulator of calcium and phosphate metabolism in bone and kidney. EB613 is an oral formulation of synthetic hPTH (1-34), (teriparatide), a peptide consisting of the first 34 amino acids of PTH which represent the functional region. Subcutaneous Forteo® (teriparatide injection) has been the leading anabolic treatment of osteoporosis since 2002. EB613 utilizes Entera’s oral drug delivery platform which promotes enteric absorption and stabilizes teriparatide in the gastrointestinal tract. Entera’s Oral PTH formulations have been administered collectively to a total of 225 subjects in two Phase 1 studies and 3 phase 2 studies (including 35 in 2 phase 2 hypoparathyroidism studies). The most recent study was a dose ranging Phase 2 study in postmenopausal women with low bone mass. This study met primary and key secondary endpoints and was presented in a late-breaker oral presentation at the ASBMR 2021 conference. For the primary efficacy endpoint: a statistically significant increase in P1NP (a bone formation marker) at 3 months was achieved. A significant dose response was observed for 0.5, 1.0, 1.5 and 2.5 mg oral PTH doses on P1NP, Osteocalcin and bone mineral density (BMD). Subjects receiving the 2.5 mg dose of EB613 showed significant increases in dose-related BMD at the lumbar spine, total hip, and femoral neck at 6 months. Subjects receiving the 2.5 mg dose of EB613 daily for 6 months had a significant placebo adjusted increase of 3.78% in lumbar spine BMD (p<0.008) which is similar to the 3.9% increase in lumbar spine BMD seen with Forteo® in clinical studies reported in the literature. Increases in total hip and femoral neck BMD were greater than those previously reported with Forteo®. EB613 exhibited an excellent safety profile, with no drug related serious adverse events. The most common adverse events included mild nausea, moderate back pain, moderate headache, and moderate upper abdominal pain.

‒Study describes a dual mechanism of action approach to oral drug delivery of large molecules, the “Holy Grail” of drug development.

‒Technology is utilized in Entera’s EB613 oral PTH drug for osteoporosis and several other molecules now in development by Entera and in collaboration with pharmaceutical companies.

BOSTON and JERUSALEM, Oct. 28, 2021 (GLOBE NEWSWIRE) — Entera Bio Ltd. (NASDAQ: ENTX), today announced that researchers at the company and the University of East Anglia in the UK published preclinical in vivo studies which demonstrate the technologies efficiency in the oral delivery of large molecule biologic drugs. The approach described in the research underpins Entera’s lead product, EB613, an oral parathyroid hormone (hPTH) entering Phase 3 clinical development for the treatment of osteoporosis. The pivotal Phase 3 study is planned for 2022.

The studies were published on line in the peer-reviewed International Journal of Pharmaceutics X.

“Entera’s highly innovative technology has the potential to transform the way we treat many diseases,” stated William Fraser, MD, Professor of Medicine at The Norfolk and Norwich University Hospitals and Norwich Medical School at the University of East Anglia. “Not only can replacing injections improve patient compliance and reduce patient discomfort, but the administration of oral PTH in both hypoparathyroidism and osteoporosis will allow for the customized titration of dose so critical to each individual patient. At present, most injections are administered via “pen” injectors, which are frequently not amenable to dose titration.”

“Entera’s innovation is to combine in a single formulation two approaches to enhancing the delivery of large molecule biologics,” said Spiros Jamas, the company’s CEO. “This research supports our strategy to address the two key challenges in delivering hPTH to the bloodstream at appropriate therapeutic levels consistently: protecting proteins from breaking down in the gastrointestinal tract; and 2) improving absorption of proteins through the gastro-intestinal tract.”

“With a burgeoning number of injectable biologics on the market and in development, there is a growing need for effective oral formulations especially in chronic conditions where replacing injections with oral dosage may result in higher patient compliance, adherence, and quality of life,” stated Entera CEO Spiros Jamas. “Beyond the near-term opportunity for our EB613 oral PTH in osteoporosis, Entera’s platform technology has applications in many other indications currently treated through injectables.”

In this paper researchers evaluated three different formulations of oral hPTH (1-34): 1. hPTH with a protease inhibitor which can protect the molecule in the gastrointestinal tract, and 2. hPTH with just a permeation enhancer (SNAC) which allows for the trans-cellular absorption of large molecules in the GI tract, and 3. hPTH combined with both protease inhibitor and permeation enhancer (the formulation technology currently utilized by Entera). In studies utilizing a porcine model, the Entera combined formulation achieved significantly higher PTH blood levels (Cmax and AUC, ~10- and ~20-fold respectively) as compared to each of these approaches on their own. The researchers concluded that an appropriate combination of these different technological approaches considerably contributes to the efficient oral delivery of a number of biological macromolecules.

The study is titled, “The combined effect of permeation enhancement and proteolysis inhibition on the systemic exposure of orally administrated peptides: Salcaprozate sodium, soybean trypsin inhibitor, and teriparatide study in pigs.” The authors on the paper are, Gregory Burshtein, Constantin Itin, Hillel Galitzer and Phillip Schwartz, from Entera Bio, Jonathan C. W. Tang, of the University of East Anglia (Norwich, UK) William D. Fraser, of the University of East Anglia and Norfolk and Norwich University Hospital (Norwich, UK).

Linear Dose Responses Seen in Biomarker and Bone Mineral Density Data

Pivotal Phase 3 registration study expected to commence 2022

BOSTON and JERUSALEM, Oct. 06, 2021 (GLOBE NEWSWIRE) — Entera Bio Ltd. (NASDAQ: ENTX), a leader in the development of orally delivered large molecule therapeutics, presented the 6-month bone mineral density (BMD) data from its completed Phase 2 clinical trial of EB613, an oral formulation of human parathyroid hormone (1-34), or PTH, for the treatment of osteoporosis. The Late Breaking Presentation “A Six-month Phase 2 Study of Oral PTH in Postmenopausal Women with Low Bone Mass – 6 Month Bone Mineral Density (BMD) Results” was selected for a prestigious oral presentation, given by Entera’s Chief Medical Officer, Dr. Arthur Santora. The oral presentation was given at 12:15 PM PDT at the American Society for Bone and Mineral Research (ASBMR) Annual Meeting in San Diego on Monday, October 4, 2021. Many scientific leaders in the field of Osteoporosis expressed great excitement with the data and the possibility of an oral osteoanabolic (bone building) agent which elderly osteoporosis patients would be willing to take.

This Phase 2 Dose Ranging study in Osteoporosis showed clinically significant changes in increases in BMD at the spine, femoral neck, and total hip. These BMD changes were associated with an increase in P1NP and a decrease in the serum CTX biomarker. A difference between the increase in P1NP, a marker of bone formation and the change in CTX, a marker of bone resorption, usually indicates an increase in bone mass, and is sometimes referred to as an “anabolic window.”

Key secondary endpoints for the trial included changes in lumbar spine, femoral neck and total hip BMD, the bone markers P1NP, osteocalcin and serum CTX after 6 months of treatment.

The most important secondary objective in the study was the demonstration of a statistically significant increase in BMD of the lumbar spine. Patients receiving 2.5 mg oral PTH for 6 months had a placebo-adjusted increase of 3.78% in lumbar spine (LS) BMD (p<0.002). Additionally, there was a highly statistically significant dose dependent increase in LS BMD across all dose groups (p<0.0001). In previously published studies with subcutaneous injectable PTH(1-34) (Forteo®) no significant increases in BMD of the total hip and or the femoral neck were generally observed at 6 months. In contrast, EB613 showed dose-dependent increases in BMD at the total hip and femoral neck at 6 months, with the greatest increases in BMD observed in the 2.5 mg group. The increase in BMD at these sites has great clinical relevance as a high percent of osteoporotic fractures occur at the hip.

“We are really excited by these full 6-month BMD data which show that our oral PTH 1-34 (EB613) for the treatment of osteoporosis produced results comparable with results observed in previously published studies utilizing injectable PTH 1-34 (Forteo®),” stated Entera CEO Dr. Spiros Jamas. “The data provides an excellent foundation for our Phase 3 registration trial, which we expect to commence in 2022.” He noted that there is an enormous clinical need for an oral agent to treat osteoporosis which can build bone and thereby reverse some of the decrease in bone strength caused by Osteoporosis. EB613 addresses this need.

Dr. Santora noted, “In this study, EB613 demonstrated reliable, consistent improvements in spine and hip BMD, overcoming a major challenge of earlier efforts to develop oral PTH agents. Additionally, EB613 performed comparably to injectable PTH 1-34 for increases in lumbar spine BMD, and also showed significant increases of BMD at the femoral neck and total hip. Change in BMD is the efficacy endpoint required for a 505(b)(2) approval pathway. Interestingly, a decrease in CTX, a marker of bone resorption, was robust and continued through six months. This contrasts with the large CTX increase typically observed with injectable formulations of PTH 1-34 currently available and may reflect a favorably lower bone remodeling rate with EB613.”

The safety profile of EB613 was consistent with the known profile of subcutaneous PTH. The most common drug-related AEs were headache, nausea, dizziness and presyncope; all drug-related adverse events were either mild or moderate, and there were no serious drug-related AE. There were no treatment-emergent hypercalcemia AEs.

Osteoporosis, characterized by low bone mass and deterioration of bone tissue, leads to decreased bone strength and increased risk of fracture. EB613, an oral PTH, may address a major unmet clinical need for formulations that may facilitate treatment in high-risk patients who may not be willing to use daily or monthly injectable osteoanabolic therapeutics.

Phase 2 Six-Month Study Design and Results

Entera’s Phase 2, 6-month, dose-ranging, placebo-controlled study evaluated EB613 in 161 postmenopausal women with low BMD. Subjects in the 2.5 mg group were either treated with 2.5 mg from Day 1 or titrated to 2.5 mg over 2 months. The primary endpoint of change in P1NP at 3 months was met and previously announced by Entera. The key secondary efficacy objective was the change in lumbar spine BMD vs. placebo at 6 months. Changes in biomarkers of bone turnover and change in proximal femoral BMD were evaluated through 6 months.

Six-month BMD and biomarker data are summarized below:

  • Significant and dose-dependent increases in BMD of the lower spine, total hip, and femoral neck at 6 months (all p <0.01), with the greatest increases in the 2.5 mg EB613 treated group
  • Subjects receiving 2.5 mg EB613 for 6 months had a placebo-adjusted increase of 3.78% in lower spine BMD (p<0.008); this group combined with the titrated 2.5 mg group showed lower spine BMD increase of 2.73% (p<0.002)
  • Pooled 2.5 mg EB613 group had significant placebo-adjusted 2.76% increase in femoral neck (p<0.002) and a 1.84% increase in total hip BMD (p<0.02) at 6 months. Significant decrease in serum CTX (of 21% from baseline at 6 months (P <0.01) while P1NP was unchanged
  • The safety profile of EB613 was consistent with and similar to the known profile of injectable PTH (Forteo®)

BOSTON and JERUSALEM, Aug. 02, 2021 (GLOBE NEWSWIRE) — Entera Bio Ltd. (NASDAQ: ENTX), a leader in the development of orally delivered large molecule therapeutics, today announced its poster presentation titled “A Six-month Phase 2 Study of Oral PTH in Postmenopausal Women with Low Bone Mass – An Interim Three-Month Analysis” has been selected by the Program Committee of the American Society for Bone and Mineral Research (ASBMR) for its Annual Meeting scheduled to take place on October 1-4, 2021 in San Diego, California. Dr. Arthur Santora, Entera’s Chief Medical Officer, will present the poster at both the plenary session on October 1 and the general session on October 2, as well as a special session on the Biology of the Aging Skeleton Symposium on September 30.

The Phase 2 clinical trial of EB613, an oral formulation of human parathyroid hormone (PTH) (1-34), for the treatment of osteoporosis met its primary and key secondary endpoints, positioning it to be the first oral bone building (anabolic) product to treat osteoporosis patients. The full 3-month biomarker data will be presented at the ASBMR annual meeting. Three and 6 month results, previously reported by Entera in June 2021 can be found here.

Osteoporosis, characterized by low bone mass and deterioration of bone tissue, can lead to decreased bone strength and increased risk of fracture. An injectable form of PTH(1-34) currently on the market, Forteo® has been shown to reduce vertebral fractures by 65-80%. However, because injections deter many patients from receiving treatments, oral EB613 may address this unmet clinical need by reducing fractures at a similar rate.

“We look forward to presenting EB613’s Phase 2 data to the leading physicians, researchers, and clinical investigators in the field of bone metabolism and health. The presentation, selected for multiple sessions, is recognition of the importance of our results and well-timed as we prepare for EB613’s pivotal Phase 3 head-to-head study vs. Forteo which we expect to initiate in 2022,” stated Entera CEO Spiros Jamas.

  • BOSTON and JERUSALEM, June 23, 2021 (GLOBE NEWSWIRE) — Entera Bio Ltd. (NASDAQ: ENTX), a leader in the development of orally delivered large molecule and biologic therapeutics, announced the final 6-month bone mineral density (BMD) results from the completed Phase 2 clinical trial of EB613 for the treatment of osteoporosis. EB613 is an oral formulation of human parathyroid hormone (1-34), or PTH, positioned to be the first oral bone building (anabolic) product to treat osteoporosis patients. Currently, fewer than 5% of osteoporosis patients on any form of therapy are treated with an injectable anabolic agent, widely accepted as the most effective form of treatment1. The Phase 2 clinical trial of EB613 was a 6-month double blind, dose-ranging, placebo-controlled study in 161 postmenopausal female subjects with osteoporosis, or with low bone mineral density (BMD). This study was conducted at four leading medical centers in Israel to evaluate the safety and efficacy of varying doses of EB613. All lab tests including biomarkers and safety monitoring were performed at a certified central laboratory, and BMD data from clinical sites was analyzed at an independent certified global imaging center.

    The most important BMD endpoint — change in lumbar spine (LS) BMD after 6 months — was met. There were statistically significant dose-related trends in the increases in LS BMD as well as femoral neck and total hip BMD, with the largest increases observed in subjects treated with EB613 2.5 mg. Dose dependent increases in biochemical markers of bone formation were previously reported. A significant increase in lumbar spine (LS) BMD was observed in the 1.5 mg group, the non-titrated 2.5 mg group (those who received 2.5 mg for the full 6 months) and the titrated 2.5 mg group (who received lower doses during titration and 2.5 mg for 4 months). An increase in LS BMD is the primary endpoint for the 505(b)(2) pathway as was described by the FDA in Entera’s pre-IND meeting. At present it is believed that the single Phase 3 Pivotal study necessary under the 505b2 pathway would require a 12-month head-to-head study against Forteo® (the “reference drug”), designed to achieve non inferiority for increase in BMD of the lumbar spine.

    Increases in LS BMD versus placebo observed at 6 months in previous Forteo® studies conducted with similar patient populations, were in the 3.9% range2. In the current study LS BMD increased 3.78% (p<0.008) in the group treated with 2.5 mg for the full 6 months. When this group was combined with the titrated 2.5 mg group (who received lower doses during titration and 2.5 mg for just 4 months) LS spine BMD increased, 2.73% (p<0.002).

    Furthermore, EB613 had a significant impact on both femoral neck and total hip BMD at 6 months. The 2.5 mg EB613 treatment group had a 2.76% (p<0.002) increase in femoral neck, and a 1.84% (p<0.02) increase in total hip at 6 months, as compared to placebo. In contrast, significant increases in BMD of the femoral neck and total hip are usually not observed with Forteo® treatment at 6 months. Increases in hip BMD have been shown to correlate with decreases in non-vertebral fracture risk3.

    In this dose-ranging study, various doses of EB613 were tested for their effect on markers of bone metabolism after 3 months and BMD of the lumbar spine, femoral neck, and total hip after 6 months. Subjects were initially randomized to receive oral EB613 0.5 mg, 1.0 mg, 1.5 mg, or matching placebo once daily. The study utilized an adaptive design with a limited interim analysis of 3-month biomarker changes in the first 80 subjects treated that demonstrated significant, dose-related increases in P1NP (a bone formation marker) after 1 month of treatment. Based on the analysis of the interim data the 2.5 mg dose was introduced.

    As previously reported, the trial’s primary endpoint was met – the complete 3-month results from the trial showed a significant increase in the P1NP biomarker in the 2.5 mg dose group after 3 months of treatment (P <0.04) as compared to placebo. P1NP is a biomarker that indicates the rate of new bone formation.

    Secondary endpoints in the trial included the effect of treatment on several additional serum bone biomarkers at 3 months including, Osteocalcin and CTX. Similar to P1NP, Osteocalcin is a biomarker for bone formation by osteoblasts, the cells that build new bone. CTX is a biomarker that indicates the rate of bone resorption by osteoclasts, the cells that remove old bone. An osteoanabolic, or bone building effect, is based on the difference in bone formation and bone resorption. An increase in P1NP or Osteocalcin, for example, associated with a smaller increase (or decrease) in CTX, usually results in an increase in bone mass.

    The decrease in CTX taken together with the increase in P1NP and Osteocalcin would indicate a potential positive impact on BMD and a reduced risk of fractures, which is the goal of an anabolic osteoporosis treatment, as reflected in the 6-month BMD results.

    The study medication, EB613, was generally well tolerated throughout the 6 months of treatment. There were no adverse events that were severe in intensity in any treatment group and no serious drug-related adverse events. However, subjects randomized to the 2.5 mg dose of EB613 presented a higher rate of adverse events (AEs), which are in line with AEs known to be associated with daily injections of PTH, such as nausea, headaches, and dizziness (or presyncope). In the clinical study setting, with subjects who are not severely osteoporotic and the COVID pandemic resulting in greater hesitation to remain in a clinical study, some of these expected AEs were resulting in subjects withdrawing their consent. Exploiting one of the advantages of an oral treatment, a novel titration regimen was introduced through a protocol amendment. Subjects randomized to the 2.5 mg dose (or matched placebo) initiated their treatment with a 1.5 mg dose followed by a 2.0 mg dose at their month 1 clinic visit and ultimately starting the 2.5 mg dose at the Month 3 clinic visit. This titration regimen minimized adverse events resulting in subjects’ drop-outs, which were within the projected rate of 20% overall, despite the COVID-19 Pandemic.

    “We are very excited and encouraged by these great results which will support advancing discussions with potential strategic partners. These results are in line with our previously reported biomarker results and further validate Entera’s platform technology and its potential to enable oral formulation of various large molecules for a range of indications that could benefit from an oral drug,” said Spiros Jamas, CEO of Entera Bio. “We are looking forward to an end of Phase 2 meeting with the FDA. More detailed results will also be presented in a future scientific conference and publications. The company will evaluate potential additional osteoporosis market opportunities specifically related to increases in hip BMD.”

    About EB613

    EB613 is an orally delivered human parathyroid hormone (1-34), or PTH, drug candidate positioned as the first potential once daily, oral, bone building (anabolic) treatment for osteoporosis patients. Teriparatide for injection (marketed under the brand name Forteo®) was approved in the U.S. in 2002 for the treatment of osteoporosis in men and postmenopausal women who are at high risk for having a fracture and is taken daily via a subcutaneous injection.

    About Osteoporosis

    Osteoporosis is a disease characterized by low bone mass and structural deterioration of bone tissue, which leads to greater fragility and an increase in fracture risk. Osteoporosis is also a silent disease, often displaying no signs or symptoms until a fracture occurs, leaving the majority of patients undiagnosed and untreated, representing a high unmet medical need. The debilitating effects of osteoporosis have substantial costs and osteoporotic fractures create a significant healthcare burden. An estimated two million osteoporotic fractures occur annually in the United States, and this number is projected to grow to three million by 2025. The National Osteoporosis Foundation (NOF) has estimated that eight million women already have osteoporosis, and another approximately 44 million may have low bone mass placing them at increased risk for osteoporosis. In US women 55 years of age and older, the hospitalization burden of osteoporotic fractures and population facility-related hospital cost is greater than that of myocardial infarction, stroke, or breast cancer.

    BOSTON and JERUSALEM, April 06, 2021 (GLOBE NEWSWIRE) — Entera Bio Ltd. (NASDAQ: ENTX), a leader in the development of orally delivered large molecule therapeutics, today announced the Company will deliver a poster presentation titled “Pharmacokinetics of an Oral Human Growth Hormone (hGH) Formulation in Rats and Mice” at the 31st Annual European Pharma Congress in London on April 28-29, 2021.

    Prescription human growth hormone (hGH), a widely used therapeutic molecule, is currently only administered via subcutaneous injection for the treatment of growth hormone deficiency as well as other indications. Therapeutic hGH was a $3.7 billion market in 2020 and is expected to grow to $8.5 billion by 2027. Also known as somatotropin, therapeutic hGH is different from oral over-the-counter supplements which claim to increase the body’s own production of hGH.

    In a preclinical study, Entera’s hGH formulation was administered orally to mice and rats. Plasma samples analyzed showed substantial gastrointestinal absorption of the oral hGH formulation and significant systemic exposure to the drug.

    “Our proprietary oral protein delivery platform continues to present expanding therapeutic applications and corresponding market opportunities. An oral hGH may offer numerous advantages including greater patient compliance, reduced pain, longer shelf life, no injection site reactions, and lower immunogenicity,” stated Entera CEO Spiros Jamas. “We are pleased to have our abstract accepted by the European Pharma Congress and look forward to presenting the data.”

    ‒ Trial Met Primary Endpoint of Significant Increase in P1NP Based on Final Analysis of 3 Month Data ‒
    ‒ Final Analysis of 6-Month Data, Including Change in Bone Mineral Density, Expected in Q2:21 ‒

    BOSTON and JERUSALEM, March 11, 2021 (GLOBE NEWSWIRE) — Entera Bio Ltd. (NASDAQ: ENTX), a leader in the development of orally delivered large molecule therapeutics, announced the complete 3-month bone biomarker data analysis from the ongoing Phase 2 clinical trial of EB613. EB613 is an orally delivered human parathyroid hormone (1-34), or PTH, positioned to be the first oral bone building (anabolic) product to treat osteoporosis patients. The Phase 2 clinical trial of EB613 is a 6-month, dose-ranging, placebo-controlled, study in postmenopausal female subjects with osteoporosis, or low bone mineral density (BMD), and is being conducted at four leading medical centers in Israel to evaluate the safety of EB613 and identify the best dose(s) for a potential Phase 3 registration trial.

    The trial’s primary endpoint was met – the complete 3-month results from the trial showed a significant increase in the P1NP biomarker in the 2.5 mg dose group after 3 months of treatment (P <0.04) as compared to placebo. P1NP is a biomarker that indicates the rate of new bone formation and the change at 3-months is the primary endpoint in this Phase 2 trial.

    Secondary endpoints in the trial comprised the effect of treatment on several additional serum bone biomarkers at 3-months including, Osteocalcin and CTX. An increase in P1NP or Osteocalcin, for example, associated with a smaller increase (or decrease) in CTX, usually indicates an increase in bone mass.

    Similar to the increase in P1NP, a significant increase in Osteocalcin was also observed in the 2.5 mg group after 3 months (P <0.01). In line with a potential anabolic effect, a significant decrease in CTX was observed after 3 months of treatment (P <0.015). The decrease in CTX taken together with the increase in P1NP and Osteocalcin would indicate a potential positive impact on BMDand a reduced risk of fractures, which is the goal of an anabolic osteoporosis treatment.

    Biomarker data from the Placebo and EB613 2.5mg dose group are summarized below:

    • A significant increase in P1NP from baseline versus placebo at month 3 (P <0.04) as well as significant increases at months 1 (P <0.0001) and 2 (P <0.003);
    • A significant increase in Osteocalcin from baseline versus placebo at month 3 (P<0.006) as well as significant increases at months 1 (P<0.0001) and 2 (P<0.0001);
    • A significant decrease in CTX from baseline versus placebo at month 3 (P <0.015) as well as a significant decrease at month 1 (P <0.001)

    Study Medication, EB613 or placebo, was generally well tolerated through 3 months of treatment. Common adverse events resembled those known to be associated with teriparatide by subcutaneous injection including dizziness, headache, palpitations, and nausea. There were no adverse events that were severe in intensity in any treatment group and no serious drug-related adverse events. Complete safety evaluations of the fully unblinded data will be conducted with the full 6-month data analyses.

    The interim analysis of the 3-month data from the first 50% (n=80) of subjects randomized demonstrated a significant increase in P1NP after one month of treatment at the 1.5 mg dose compared to placebo. The complete 3-month biomarker results now show a significant increase in P1NP at months 1 and 2 and significant increases in Osteocalcin from baseline versus placebo at months 1, 2 and 3, with the 1.5 mg dose. The complete 3-month analysis shows a significant (P<0.0001) dose response effect of EB613 (0.5, 1.0, 1.5 and 2.5 mg) on P1NP. Entera intends to submit the full dataset for publication and/or presentation at an upcoming medical conference.

    “We are very pleased and highly encouraged by the complete 3-month biomarker data from this trial. When taken together with the previously reported interim BMD data, these results support a dose response in BMD. We look forward to reporting the final results, including the 6-month BMD data, from this trial in the second quarter of 2021,” stated Entera CEO Spiros Jamas. “We believe these data support ongoing business development discussions with strategic partners with interests in osteoporosis. We also believe these data and previously reported data illustrate the power of Entera’s drug delivery platform for oral dosing of large biological molecules.”

    ‒ EB612 When Added to Standard of Care Led to a Statistically Significant Decrease in Supplemental Calcium Usage –

    ‒ Oral Human Parathyroid Hormone (1-34) Has the Potential to Have a Major Impact on Compliance, Adherence, Therapeutic Impact and Quality of Life for Patients –

    BOSTON and JERUSALEM, Feb. 25, 2021 (GLOBE NEWSWIRE) — Entera Bio Ltd. (NASDAQ: ENTX), a leader in the development of orally delivered large molecule therapeutics, announced today the publication of the results of its previously completed Phase 2a study of EB612 in the Journal of Bone and Mineral Research. The article, titled “Safety and Efficacy of Oral Human Parathyroid Hormone (1-34) in Hypoparathyroidism: An Open-Label Study,” discussed the results of the four-month study in which EB612 was evaluated in 2015 as an adjunct to standard calcium and vitamin D supplement treatment in patients with hypoparathyroidism (HypoPT). EB612, an oral human parathyroid hormone (1-34) (PTH), has received Orphan Drug designation from the U.S. Food and Drug Administration and the European Medicines Agency for the treatment of HypoPT.

    The Phase 2a study demonstrated the safety and tolerability of EB612 administered four times daily for 16 weeks to patients with HypoPT. The study achieved its primary and secondary endpoints, including a reduction in calcium supplements, reductions in serum phosphate and 24-hour urine calcium excretion, maintenance of albumin-adjusted serum calcium (ACa) within the reference range, and an improvement in quality of life. Specific results of this trial included:

    • A significant reduction of 42% (p=0.001) from baseline in median calcium supplement use;
    • Maintenance of median ACa levels above the lower target level for HypoPT patients (>7.5 mg/dL) throughout the study;
    • A rapid decline of 23% (p=0.0003) in median serum phosphate levels 2 hours following the first dose that was maintained within the normal range for the duration of the study;
    • A notable median decrease of 21% (p=0.07) in 24-hour urine calcium excretion between the first and last treatment days; and
    • An increase in quality of life score of 5% (p=0.03) from baseline by the end of the treatment period.

    In this study, patients were titrated up to a maximum of 12 EB612 0.75 mg tablets a day (total daily dose of 9 mg) by the investigator, according to each subject’s ACa, and supplement treatment regimen. Of the 19 enrolled subjects, 17 completed the trial (of which 15 were per protocol). No drug-related serious adverse events were reported and most of the adverse events were not considered study drug-related.

    “The publication of our Phase 2a EB612 study results in this leading peer-reviewed journal support our HypoPT development program and the value of our platform technology. The availability of an oral PTH is expected to improve compliance, as well as therapeutic impact and may offer patients with HypoPT a much-needed alternative to the currently available parathyroid hormone replacement therapy options which are administered via daily injections. We are currently working on improved formulations of EB612 and the design of the next clinical trial which we expect to initiate in 2022,” stated Entera CEO Spiros Jamas. “In addition, through the use of our previously announced at-the-market equity program, we have continued to strengthen our balance sheet in the first quarter of 2021, and we are now funded into the fourth quarter of 2021.”

    The full peer-reviewed publication will be available on Entera’s website once the final article is released for publication and can be found here.

    About Hypoparathyroidism

    Hypoparathyroidism (HypoPT) is a rare condition in which the body produces insufficient amounts of parathyroid hormone. Individuals with HypoPT typically exhibit abnormally low levels of calcium in the blood (hypocalcemia), and high levels of phosphate in the blood (hyperphosphatemia). They also develop increased urine calcium (hypercalciuria). HypoPT is estimated to affect approximately 77,000 individuals in the United States. Historically, the treatments for HypoPT have been calcium supplements and active vitamin D (calcitriol or alfacalcidol). Phosphate binders that inhibit phosphate absorption and thiazide diuretics that reduce urine calcium are occasionally added. It is often difficult to titrate the dose of both calcium supplements and active vitamin D to reduce symptoms of hypocalcemia without producing increased urine calcium or hypercalcemia with tissue calcification during chronic use, which can result in kidney injury and significant healthcare costs. Moreover, the high doses of calcium supplements may produce stomach and gastrointestinal symptoms as well as other symptoms that negatively affect a patient’s quality of life. A once-daily, injectable form of parathyroid hormone has been approved by the FDA and EMA for the treatment of hypocalcemia in patients with HypoPT.

    About Entera Bio

    Entera is a leader in the development of orally delivered large molecule therapeutics for use in areas with significant unmet medical need where adoption of injectable therapies is limited due to cost, convenience and compliance challenges for patients. The Company’s proprietary, oral drug delivery technology is designed to address the technical challenges of poor absorption, high variability, and the inability to deliver large molecules to the targeted location in the body through the use of a synthetic absorption enhancer to facilitate the absorption of large molecules, and protease inhibitors to prevent enzymatic degradation and support delivery to targeted tissues. The Company’s most advanced product candidates, EB613 for the treatment of osteoporosis and EB612 for the treatment of hypoparathyroidism are in Phase 2 clinical development. Entera also licenses its technology to biopharmaceutical companies for use with their proprietary compounds and, to date, has established a collaboration with Amgen Inc.
